In 2023, Ozempic (semaglutide) became the most searched drug on the planet. Not for the diabetes it was designed for, but for weight loss. What followed was predictable: millions of people searching for cheaper alternatives, without a prescription and without an injection. "Natural GLP-1," "berberine natural Ozempic," "GLP-1 booster." Google Trends shows an 800% increase in berberine searches in a GLP-1 context in the US alone.

The real question is not whether a "natural Ozempic" exists. It does not. Nothing OTC replicates the effect of semaglutide or tirzepatide. The correct question is: are there natural compounds that activate the GLP-1 pathway in a modest but measurable way? The answer is yes. But with nuances that most supplement brands do not tell you.

What GLP-1 is and why it matters

The hormone

GLP-1 (glucagon-like peptide-1) is an incretin hormone produced by L cells in the gut when you eat. Its function:

  1. Stimulates insulin secretion (only when glucose is high, unlike sulfonylureas)
  2. Suppresses glucagon (reduces hepatic glucose production)
  3. Delays gastric emptying (you feel full longer)
  4. Reduces appetite centrally (acts on the hypothalamus)
  5. Protects pancreatic beta cells (anti-apoptotic)

Honesty: calling berberine "natural Ozempic" is like calling a candle "natural sun." They act on the same pathway, but the magnitude of the effect is not comparable.

The compounds that activate GLP-1

1. Berberine: the most studied

Berberine is an alkaloid extracted from several plants (Berberis vulgaris, Coptis chinensis, Phellodendron amurense). It is the natural compound with the most evidence for GLP-1 modulation.

Zhang et al., 2008 (Metabolism): In vivo and in vitro study. 5 weeks of berberine increased glucose-stimulated GLP-1 secretion. In vitro, berberine stimulated GLP-1 release in NCI-H716 cells in a dose-dependent manner. The mechanism involves the PKC-dependent pathway.

Wang et al., 2024 (Frontiers in Pharmacology): Berberine metabolites directly stimulate GLP-1 secretion in intestinal L cells. It is not only intact berberine that acts: its metabolites produced by the gut microbiota also activate the pathway.

Li et al., 2023: Berberine improves pancreatic beta cell function through the GLP-1/GLP-1R/PKA pathway, acting on both intestinal L cells and islet alpha cells.

Glucose meta-analysis: A meta-analysis of 37 studies (3,048 patients) confirmed that berberine reduces fasting glucose, HbA1c, and insulin resistance in type 2 diabetics. Efficacy comparable to metformin in some studies.

Dosage: 500mg, 2-3 times per day (1000-1500mg total), with meals.

2. Curcumin: stimulates GLP-1 directly

Takikawa et al., 2013 (Biochemical and Biophysical Research Communications): Curcumin stimulates GLP-1 secretion in GLUTag cells through the Ca2+/calmodulin-dependent kinase II (CaMKII) pathway. The effect is direct on intestinal L cells.

Kato et al., 2017 (Journal of Nutritional Biochemistry): Curcumin improved glucose tolerance via GLP-1 secretion stimulation in mice. The mechanism: activation of GPR40/120 in L cells.

Li et al., 2023 (Nutrients): Curcumin compensates for GLP-1 deficiency through the microbiota-bile acids-TGR5/FXR axis. It acts as a natural TGR5 agonist, a receptor that stimulates GLP-1 secretion.

Limitation: curcumin bioavailability is low. You need formulas with piperine (BioPerine) or liposomal forms for adequate absorption.

Dosage: 500-1000mg of curcumin with piperine, 1-2 times per day.

3. Yerba mate: the incretin effect

Resende et al., 2025 (Nutrients): Yerba mate supplementation significantly increased GLP-1 gene expression and plasma GLP-1 levels. Dihydroferulic acid (a microbial metabolite of ferulic acid present in mate) directly stimulated GLP-1 production in L cells.

4. Soluble fiber: the physiological stimulator

Soluble fiber (psyllium, inulin, beta-glucan) ferments in the colon and produces short-chain fatty acids (SCFAs: butyrate, propionate, acetate). These SCFAs activate GPR41 and GPR43 receptors on L cells, stimulating GLP-1 secretion.

It is the most physiological mechanism: it is literally what your body does when you eat fiber. It is not a hack. It is basic nutrition that most people fail to meet (average fiber intake in the US is 15g/day vs the recommended 30g).

5. Cinnamon: modest but documented effect

Cinnamon (Cinnamomum verum) delays gastric emptying and improves insulin sensitivity. The effect on GLP-1 is indirect but contributes to the glycemic profile.

The "GLP-1 Boosters": what is inside

Kobho GLP (Spain)

Kobho Labs is the Spanish supplement brand that has grown the most in 2025-2026 (trend +800% on Google Trends Spain). Its GLP product is a booster with berberine + probiotics + chromium.

The good: Spanish brand, available on Amazon, formulation with documented ingredients. The questionable: the marketing suggests an alternative to GLP-1 medications, which is exaggerated.

ColonBroom GLP-1 Booster (US)

Trending product (+4,100% on Google Trends US). Contains psyllium fiber + probiotics + additional ingredients. The marketing angle is "GLP-1 booster" but in reality it is a fiber supplement with good branding.

  • Pregnancy: contraindicated (can cross the placenta and cause neonatal kernicterus)

Fiber

  • Safe without a practical limit (but increase gradually to avoid gas)
  • Take separately from medications (2 hours before or after, as fiber can reduce absorption)

Curcumin

  • With piperine it can increase drug bioavailability (use caution)
